1.) Between your guitar and the amp

My Mesa F-30 has a parallel loop. I put my TLSE here with the mix knob all the way up. I then control the "wetness" levels off all my effects on the TLSE. If you're planning on using the ENGL's multiple channels, the this is the route I think you should go. The problem I've found when having my delay in front of a multi-channel amp, is that I can set the delay to sound great through one of the amp's channels, but then it doesn't sound very good when I switch to the other channel. Get some good shielded cables when using your amp's FX loop; your preamp signal is traveling through those wires.
To avoid coloring the amp's tone, turn off all amp and cabinet models on the TLSE. I don't think having the TLSE in your signal chain is going to affect your tone enough to notice. Mine augments my Mesa nicely.
